In the 1980's, Greenville saw a steadily increasing number of Muslims moving into the area, as Greenville was becoming a center for engineering and technology. Up till this point in time, Greenville had no place of worship for Muslims and the community was becoming increasingly concerned about the need for Islamic education as young Muslims were growing up. Within the homes of Muslim families, classes and prayers were established and soon the increasing population of Muslims in the late 1980's would outgrow these homes. Seeing this need, several committed Muslims started to organize and come together with the goal of establishing a Masjid that would be the center of the Muslim community and education in Greenville. The Islamic Society of Greenville (ISG) was established in 1987 within a rented room in the building formerly known as "Danny's Antique Mall". This room served as our Masjid for daily and Jumma prayers. In addition, Sunday classes were held to teach the young and adult Muslims alike, the fundamentals of Islam.

Perluasan pertama dilakukan dengan wakaf Sayid Umar bin Muhammad Assegaf Altoha dan Sayid Achmad bin Syech Sahab yang dilaksanakan pada tahun 1897 dibawah pimpinan Pangeran Nataagama Karta mangala Mustafa Ibnu Raden Kamaluddin.
Perluasan kedua kali pada tahun 1930. tahun 1952 dilakukan lagi perluasan oleh Yayasan Mesjid Agung yang pada tahun 1966-1969 membangun tambahan lantai kedua sehingga luas mesjid sampai sekarang 5520 meter persegi dengan daya tampung 7.750.
